This is just my opinion, but...
Valuable: Probably a mint in box train set from the 1950's/1960's. I've seen some of those creeping up on $1000.
Rare: Pre-production samples would be the obvious. As for production samples, I'm sure it's debatable...
Unique: With Tyco where do I even start? The GG1, Turbo Trains, Talgo Streamliners, TV promotional sets, etc... The list goes on. 
BTW, you may want to pick a better sub-forum for this. This is usually where the "Accessory of the Week" threads go. You might get more responses in the "Loose Ends" section.
